CUT&Tag (Cleavage Under Targets & Tagmentation) is an epigenetic profiling method used to map DNA-protein interactions genome-wide. It uses an antibody to target a protein of interest, followed by tagmentation (cleavage and integration of sequencing adapters) by a Tn5 transposase loaded with sequencing adapters, directly at the antibody-bound site. This targeted tagmentation minimizes background and improves resolution compared to ChIP-seq. CUT&Tag is commonly used to map transcription factor binding sites, histone modifications, and other chromatin-associated proteins.
